In most cases, your employer needs a Labour Market Impact Assessment (LMIA) to support your job offer for Express Entry. A valid job offer has to be made by one employer for continuous, paid, full-time work for at least one year. Also, the job should be of Skill type O, A or B. As per your question, you need a job offer from your employer with a new positive LMIA. If you are currently working in Canada in a NOC 0, A or B job on a work permit that was issued based on an LMIA, then you do not need another LMIA.
